Abstract: A précised and reproducible HPLC method was developed for simultaneous estimation of antidiabetic drug, pioglitazone HCl and antihypertensive losartan potassium. Diabetics are prone to hypertension hence co-administration of antihypertensive and antidiabetic drugs is most common. The method is applied for their determination in pure form, in pharmaceutical formulations and in plasma sample using a Phenomenex column (100×4.6 mm, 5 μm particle size) in an isocratic mode. The mobile phase consisted of aqueous phosphate buffer (0.05 M, pH 3.5) and acetonitrile (70:30, % v/v). The detection was carried out at 225 nm. The method was linear over the concentration range 0.5–7.0 μg/ml and 0.1–7.0 μg/ml for pioglitazone HCl and losartan potassium respectively. The analytical validation of the proposed method was carried out using ICH guidelines.
